We shared the taco pizza that day, and my husband had the barbecue chicken. It was a perfectly thin pizza crust, reminiscent of an artisan oven-roasted preparation. Neither of us believed the waitress, but we took her advice that “we would never be able to tell.” Well, she made a believer of both of us. The real gem we discovered that day was the cauliflower pizza crust. At that point in our lunch, my daddy was ready to drive in from Dallas the next weekend or sometime this fall to watch the changing colors and have dinner. Our waitress kept reminding us to come back on a Thursday evening where the same portion is the weekly special of $11 while they host karaoke night and special entertainment district live music nights. The country boy across from me was not sure he would have finished it, but he wanted to try! The mounded-up mashed potatoes and green beans hung out in a moat of white gravy. The flattened beef steak was over the edges of his oval restaurant dish. My dad let me do the ordering and was very envious when the cowboy next to us had a chicken-fried steak delivered to his table. From reviews, I could see they did have a great burger, but everyone kept talking about this gigantic chicken-fried steak. When we entered the front door of The Vault, I was fully prepared to have a burger and regular drop-in-the-fryer-type appetizers. Over time, many retail businesses occupied the spot at 7th and Main until Lonnie and Mindy London opened The Vault 1905 Sports Grill. During their time, they drew timelines on the walls of historical events. While the backside was still referred to as Hinkle Hall and used for barbershops and stores throughout the years, the Press Argus Newspaper moved into the second floor, above the bank, through the 1920s, where many hand drawings still adorn the walls from the editorial room. In 1904, Citizens Bank purchased the Hinkle Corner, and John Blevins was issued a contract to build the building standing today at 7th and Main Street. As downtown Van Buren grew in artistic expression, performing groups and a Glee Club shared the property with the Masons. Let me tell you, The Vault 1905 Sports Grill is a hidden secret. After spending our morning learning about lawmen who hunted down robbers, it seemed like the perfect coupling to our day of adventures in Fort Smith. I researched using some of my usual tourism tools and found recommendations for a sports bar inside an old bank. I’ve heard much about the revitalization of the historic district of this Western Arkansas town and decided it was time for us to see it for ourselves. ![]() This, however, means the time display for the whole time you're in Geneva will be a bit counter-intuitive to read – you'll be reading the hour in Geneva off the 24-hour hand, and moreover, the date will not switch over at midnight Geneva time, but rather, six hours too late, because it will change over at midnight in New York. You can re-set the 24-hour hand to Geneva time and leave the hour and minute hand as they were, showing the time in New York. If you're traveling from New York to Geneva (today, as I write, the time difference is six hours, with Geneva ahead), when you land at GVA, you have two choices. ![]() In 1879, Scottish-born Canadian engineer Sir Sandford Fleming proposed the division of the world into 24 standard time zones, and by 1900, most countries had adopted some form of standard time, which gradually evolved into the system we know today. This time standard used GMT across the entire rail network. This meant, of course, that every town and village separated by longitude had a slightly different time, which didn't matter very much at all until the invention of the railroad – the first standard time was so-called Railway Time, which was adopted in 1840 by the Great Western Railway in the UK. Each time zone follows a standard mean time across the entire time zone prior to the development of standard times, the time was simply the local solar mean time. ![]() The multi-time-zone watch is an interesting complication – unlike many other complications, such as the moon-phase and equation of time, and even the perpetual calendar, it is not based on recurring natural events, but rather on an invented convention, which is the division of the world into time zones.

Depending on how much data you have, one could be cheaper than the other. And, while services like Dropbox are free for small amounts of space, Backblaze's low price is for as big a backup as you want. Backblaze will keep multiple copies of different versions of your files, so you can restore the file exactly as it was from many points in its history. Dropbox is designed to sync your files between PCs, while Backblaze and similar services are designed to backup large amounts of files. While backup programs like Backblaze and cloud storage services like Dropbox are both online backups, they work in fundamentally different ways. Depending on the files you want to back up, this method can either be simpler or more complicated than a straight-up backup program. Pros: This method is easy, fast, and in many cases, free, and since it's online, it protects you against all types of data loss.Ĭons: Most cloud services only offer a few gigabytes of space for free, so this only works if you have a small number of files you want to back up, or if you're willing to pay for extra storage. If your hard drive dies, you'll still have the copies of the files stored online and on your other computers. The service you choose will then automatically sync to your online account and to your other PCs. Rather than just storing your files on your computer's hard drive, you can store them on a tool like Dropbox, Google Drive, Microsoft OneDrive, or a similar cloud storage service. Use a Cloud Storage Serviceīackup purists will say this isn't technically a backup method, but for most people, it serves a similar enough purpose. All the computers on your home network can back up and restore from the NAS. You can get a NAS (network-attached storage) device instead. To create backups and store them locally, you don't have to plug an external hard drive into all the computers you use. Restoring a backup can take a while, too. Pros: Online backup protects you against any type of data loss-hard drive failure, theft, natural disasters, and everything in between.Ĭons: These services usually cost money, and the initial backup can take much longer than it would on an external drive-especially if you have a lot of files. The accuracy of TCR repertoire characterization (focusing on the short, but extremely variable, CDR3 region) has further been improved by using 5′ Rapid Amplification of cDNA Ends (5′ RACE) 15, which alleviates the amplification bias introduced by a multiplexed PCR 16, 17. However, in the past decade, high-throughput sequencing (HTS) has revolutionized and prompted direct, affordable, and efficient sequencing of immune repertoires 13, 14. Consequently, profiling of immune repertoires was limited to a few model species of medical or veterinary importance. Indirect techniques, such as CDR3 spectratyping 11, allowed a general overview of TCR repertoire dynamics, but a direct exploration of sequence diversity was only possible with exceedingly time-consuming and costly cloning and Sanger sequencing (reviewed in Six et al.) 12. The composition and size of the TCR repertoire is crucial for a successful immune response 8, 9, 10 however, its enormous diversity has long impeded in-depth study of an individual TCR’s collections at the genomic level. In the house mouse ( Mus musculus), the number of unique TCR αβ pairings was estimated at 1.9 × 10 6, with the number of unique TCRβ chain types estimated to reach 5–8 × 10 5 clonotypes 7. The size of an individual αβ TCR repertoire of mature lymphocytes has still not been precisely determined even in humans, with estimates differing by an order of magnitude 5, 6 from 2.5 × 10 7 to 1 × 10 8. The functional TCR repertoire is shaped during maturation in the thymus, where processes of positive and negative selection assure MHC restriction and central tolerance of T cells, respectively (reviewed in Klein et al.) 4. As a result, each new T cell is provided with a potentially unique receptor, generating an extremely diverse TCR repertoire 2. CDR3 encompasses the V(D)J junction, where insertion and/or deletion of random nucleotides between the rearranged segments (N-diversity regions) increases diversity over that introduced by recombination between V, D, and J genes. The most variable fragment of a TCR – the Complementarity Determining Region 3 (CDR3) – is responsible for recognition of the peptide–MHC complex. In the murine genome, there are approximately 100 V and 60 J segments of α chain, and 35 V, 12 J, and 2 D segments of the β chain however, up to one third of them are pseudogenes or otherwise non-functional 3. During somatic recombination, V (variable) and J (joining) segments form the variable domain of the α chain, whereas the equivalent domain of the β chain is composed of the recombination of three segments: V, J, and D (diversity). The variability of TCRs, necessary to ascertain specificity and robustness for interaction with different antigens, is generated similarly to that of immunoglobulins–that is, by a random, somatic recombination of different, germline DNA segments (Fig. The main function of αβ TCRs is recognition of foreign peptides in the context of the major histocompatibility complex (MHC) 2. In general, majority of circulating T cells bear receptors formed by α and β chains (however this varies between tissues and species, with frequencies ranging from 90–99% in human, mice or rat to 60–85% in some ruminants, pigs or chickens) 1. The central role of T cells in the immune response, both humoral and cytotoxic, is mediated by membrane, heterodimeric T-cell receptors (TCRs). comparative immunology, ecology and evolutionary biology. We hope that by providing an easy-to-follow molecular protocol and on-line bioinformatics tools that do not require reference sequences (AmpliTCR and AmpliCDR3), we will encourage HTS immune repertoire profiling in other non-model vertebrates, thus opening new research avenues in e.g. ![]() A conservative, lower bound estimation of the TCRβ repertoire was similar to that found for mice (1.7–2.3 × 10 5 clonotypes). Further, we implemented unique molecular identifiers for quantitative analysis of CDR3 repertoire with stringent error correction. Overall orthology to murine genes was preserved, with 11 J and 37 V genes found in voles (although 3 V genes lacked a close orthologue). ![]() We used RNA from spleens, 5′RACE and HTS to describe V and J segments of TCRβ, qualitatively characterize preferential V–J segment usage and CDR3 length distribution. We present the first in-depth characterization of the T-cell receptor (TCR) repertoire in a non-model mammal (bank vole, Myodes glareolus), widely used in ecological and evolutionary research. However, fast progress in the field applied mostly to human and mouse research, with only few studies devoted to other model vertebrates. In recent years, immune repertoire profiling with high-throughput sequencing (HTS) has advanced our understanding of adaptive immunity. ![]() Since data science model training is based on simple matrix operations, GPUs can be used safely for deep learning.
